Replaced the batteries twice with new Duracell’s. The unit is back online, and the valves work, no moisture, but we still get a beep every 60 sec?

Defective Unit. Have two units for a year. So Far, So Good. Cycling each month is a good idea. Batteries are a Backup system for power outage.

Is there an indication (light etc.) if the unit is running only on batteries but not on AC power?

no. only low battery notification

If there is problem with the unit can it be opened manualy?

Yes, there are instructions in the manual included. I elected to install a parallel shut off valve beside the flood stop valve. I purchased two tees and a valve. I had some pex pipes and crimp clamps on hand, so minimal expense. Valve is normally closed. If the flood stop would close and you want water, open your bypass valve. Be careful, you may have a leak.

Do I really need this if my tank is by a couple of floor drains?

Why wouldn't you even with the drains? Your tank leaks, the valve closes, the water stops and you buy a new heater. It's like an insurance policy. Hopefully you don't use it but it's good to know it's there if you do..
