Q: Could this be used as a fishing backpack? Is it big enough for a few Plano boxes?

Yes totally I used it for work I pretty sure a laptop could fit in this backpack. You could try to fit 2-3 Plano boxes!

Q: Can I comfortably strap a machete to this?

I think you could. If the side straps can't hold it securely enough, it looks like you could slide it down the compartment on the back of it (next to space for hydropack) and zip it closed as far as u could. It being in the back, pressed against your back, it should stay put.

Q: Will this pack big enough for a 6'3" 320,lb man?

Probably... not, IMO. It depends on what you need to carry though. The bag itself will likely fit you. I bought this because I thought having the included holster would be nice. I am 6'1 and 200lbs. Although it fits and I do hike with it, it is a smaller pack. The holster itself is too small and useless, and my firearm (Sig 226, full size frame handgun) takes up the entire compartment and I ended up just ditching the holster. It's a great small pack. But it is a "small" pack as it says in the name. Overall I'm happy with it for what I use it for, short 3-6 hour hikes. It carries water, the firearm, and a small basic first aid kit. Those items have the bag full.
