Q: Does this work for speaker cable?

They should be ok providing you are not joining out more than three core and the cable is 4-8mm diameter, I used them for LED garden lighting cables.

Q: Can the back of the box be drilled for a hole ? to run a mains cable through the wall to link an outdoor socket and a 1.5mm swa mains cable to shed ?

If you drill it will no longer be IP RATED as there will be no seals, you could do this but would need a good seal around the hole you drill using something like silicon sealant so really it is doable but down to your skills in modifying and waterproofing the new hole

Q: Why 5 screw holes to seal the top but only 4 screws supplied?

No idea, but I lost one of the screws whilst assembling it for the first time so I only used four (on the corners) and it seems pretty watertight to me after 4 months. Careful though, once the membrane/washer has been punctured I reckon you would need a screw to seal the hole.
