Can this be placed on top of an already in use screen protector? Will it still magazitize?

It can place on top of the screen protector. That is the purpose of the magnet. When you do not need to write or take note anymore. Easily take it out and put it away until you need it.

How do I clean the protector? Can I wash it with water and let it dry?

Yes you can wash it with water. I clean my with a slightly wet microfiber cloth.

Does this leave enough space around the edges so as not to interfere with a plastic ipad case? (Not just a magnetic cover)

I’m currently using this with a clear plastic ipad case and I see no interference with the screen cover. It will probably depend on the case you are using though.

I bought two of them and both of them only have one side of strip has magnetification.Is that right or both of them are defected ?

There should be two magnetic strips- one on left and one on right side of screen. Both on same side of screen. When placed on iPad the magnetic strips are on front of screen. Hope that answers your question.
