Q: Would these work for kitchen cabinets and drawers

I would have to say "np" because there is only on hole for a screw which is in the center of the handle. Most kitchen cabinet handles have two screw holes.

Q: I'd like to purchase a few replacement knobs only, for color choices/scheme. is this possible?

This is probably a question for manufacturers not customers. But I have purchased these and you get one color in each set. My guess is you have to purchase a complete aet

Q: The knob doesn't seem to tighten when using the screw. We attached the item to the cabinet door front but it remains loose.

I am not sure if this is your problem but I had to buy longer screws because if the thickness of the cabinet drawers I was using these on.
