Cepham Life Sciences 50X TAE (Tris-Acetate-EDTA) Buffer contains 2M Tris, 1M Acetic acid with 0.05M EDTA and is used for running electrophoresis of nucleic acids in the gels for providing electrical conductivity and maintaining the pH. The EDTA present in the buffer inhibits metal dependent nucleases by chelating the divalent cations (Ca2+, Mg2+), and thereby protects the DNA from nucleases during the electrophoresis. The buffer may be used for both genomic and large supercoiled DNA, and also be as a gel preparation buffer. This solution has a lower buffering capacity than TBE buffer, therefore, periodic replacement of the buffer during prolonged electrophoresis is recommended. However, dsDNA runs faster with TAE buffer than TBE. The 50X concentrated TAE buffer should be diluted with molecular biology grade water to 1X before running the electrophoresis.
