Q: Hi! Would you let me know how the spray mister is supposed to be used? To spray on the blender when it's dry or directly on the paper?

You can mark on the plastic with your Tombow markers and then spray it with water and rub your paper onto the plastic for a beautiful blended "water color" look. Sprinkle with salt for an even cooler appearance (wipe the salt off your paper after it dries.)

Yes, I've been using it for watercolor pencils, any medium that says "water soluble" will work, water colored pastels also. I like em' rather then fully using those water pens, tip doesn't absorb too much, so more control of blending. But it depends what u want for ur drawing. I use both. In fact, I'm buying more, then I saw ur question. I like this more than the Prismacolor & Lyra blenders for pencils, which I also hav for othr things. They all hav various uses for dimension in drawings. It all depends what ur trying to convey in ur art.

No, I don’t think that will work. It’s not meant for fabric nor is it meant for colored pencils. It is specifically for Tombow Dual Brush Pens.

Q: Is the mister was empty

I'm not sure if that was a typo, but if you're asking if the mister comes empty it does. I fill it with water when I want to use it and empty it when it's done.
